Motor-Activity Markers of Circadian Timekeeping Are Related to Ketamine’s Rapid Antidepressant Properties

Abstract

Background

The rapid clinical antidepressant effects of the glutamatergic modulator ketamine may be due to its ability to restore synaptic plasticity and related effects on sleep-wake and circadian systems. Preclinical studies indicate that ketamine alters expression of circadian clock–associated molecules, and clinical studies of Ketamine online plasticity-related biomarkers further suggest an association with sleep slow waves and sleep homeostasis.

Methods

Wrist-activity monitors were used to examine the pharmacologic and rapid antidepressant effects of ketamine on markers of circadian timekeeping (amplitude and timing) in mood disorders. Circadian amplitude and timing of activity at baseline, postinfusion day 1 (D1), and day 3 (D3) were measured in 51 patients with major depressive disorder or bipolar disorder.

Results

Compared with either placebo or baseline, a mood-independent decrease of the central circadian value (mesor) was present on D1 after ketamine treatment. Mood-associated … Read More

Buy now, pay whenever? Lockdown lift for online shopping loans

By Nikhil Nainan

(Reuters) – Browsing online during lockdown, Jessica Friend spotted a pair of Ray-Ban sunglasses she liked, but the price tag made the 30-year-old Ohio resident think twice.

What persuaded her to click ‘buy’, Friend said, was the short-term credit offered by Afterpay, which split the $260 payment into four interest-free instalments.

Afterpay is among a handful of alternative credit firms which offer small loans, mostly to online shoppers, and make their money by charging merchants a 4%-6% commission.

These buy-now-pay-later (BNPL) firms have benefited from a shift to online shopping during the coronavirus crisis in countries including the United States, where state aid has also boosted retail sales.

“I’m more inclined to use them because they make it easier to afford to get the things I want all at once … and when I want to splurge on something,” Friend said of the loans.

Consumers Remain Wary About In-Store Shopping

Though stay-at-home orders have ended and certain retail stores have reopened, not all consumers are rushing to return to their pre-coronavirus shopping habits.

Consumer research company First Insight has been tracking shoppers’ sentiments amid the COVID-19 pandemic since late February. The most recent survey, conducted on July 10, 2020, found that many shoppers are uncomfortable with in-store interactions, and that safety concerns are rising over time in some cases.

Fashion, beauty take a hit

While consumers have been looking forward to returning to their pre-coronavirus routines, many may be finding that it’s taking longer than expected. For example, many shoppers are still afraid to try on clothes. In fact, 68% of women surveyed in July said they felt unsafe trying on clothes in dressing rooms, up slightly from 65% of women previously surveyed in April.

An even higher percentage of women (80%) noted discomfort with trying out beauty

Make Juneteenth a Non-Shopping Holiday

Juneteenth, tomorrow, has long been celebrated by Black people and across communities of color. The holiday references June 19, 1865, two-and-a-half years after the Emancipation Proclamation, when a document declaring the end of slavery was read publicly in Galveston, Tex., by a U.S. Army general. Now the day is getting broader recognition, as numerous American companies, including some in fashion, are making it a paid holiday. Congress will likely take up the issue as well, and Juneteenth could, and should, become a national holiday.

How great and long overdue for our country to formally celebrate the end of slavery, and by doing so, come face-to-face with the horrific devastation it wrought and that is ongoing. Not so great that this reckoning comes only after the killings by police of George Floyd, Breonna Taylor, Rayshard Brooks and too many others, acts that have shaken
